Prenatal exposure to NaF alters dental eruption by decrease of osteoclast number in the molar bone crypt of rats

Tooth eruption (TE) is a tightly regulated and spatially and temporally programmed process. In previous work, we demonstrated that prenatal exposure to NaF 50 mg/L produced an increase in the basal bone volume of the mandible and a decrease in the degree of tooth eruption. The aim of the study was to study the morphological and biochemical events triggered by prenatal NaF exposure during the pre-eruptive stage in the bone crypt of the first rat molar. Three- and 10-dayold offspring (n = 5) from two groups of mothers were used: controls (0.3 mg/L NaF) and treated (50 mg/L NaF). Thetreatment was performed during pregnancy and lactation. Animals were euthanized at 3 and 10 days by cervical dislocation. Plasma calcium y phosphorus and total alkaline phosphatase (FAL) were determined. Mandibles were removed and histologically processed to obtain buccolingual sections stained with H&E or histochemical detection of tartrate-resistant acid phosphatase (TRAP). In sections of first mandibular molar of 3- and 10-day-old pups, the following histomorphometric parameters were evaluated: trabecular bone volume (BV/TV [%)]) and number of osteoclasts (N.Oc/mm2 ) and TRAPþ osteoclast number. Results were expressed as mean SE and analyzed by Student’s test. Phosphatemia increased in 3- and 10- day-old pups from mothers treated with Fcompared with those from controls (p < 0.05). FAL increased in the 10-day-old pups exposed to F- . Histomorphometric analysis showed an increase in the BV/TV of the bone crypt of 3- and 10-day-old pups exposed to F- (p < 0.01); this increase was associated with a decrease of N.Oc/mm2 (p < 0.01) and the number of TRAPþ cells. In conclusion, the increased BV/TV and the lower N.Oc observed in the bone crypt of the mandible of 3- and 10-day-old pups from mothers treated with NaF suggested a disruptive effect triggered by F- on the formation events of the eruptivechannel in the offspring, which impaired the degree of eruption.
