Cenchrus ciliaris L., is an important gramineous used as forage for ruminants in north-west areas of Argentina. It reproduces mainly by aposporous apomixis. One way of obtaining a novel germplasm is by using induced physical mutations as X rays. Because of the random nature of mutagenic agents, it is necessary to use accurate techniques that enable to characterize the new genotypes. In the last decade, the inter-simple sequence repeat (ISSR) markers have been recognized as low-cost and useful molecular markers for genetic variability analysis. The aims of this work were to estimate the induced genetic variability among putative mutants by means ISSR markers and evaluate the discrimination capacity of this technique. For induce mutations, mature seeds of RN 49, RN 51 and RN 148 accessions were exposed to X rays (400 Gy). Following mutagenic treatment, fresh leaves of obtained seedlings were used to perform DNA extraction. A total of 28 primers were screened initially. The 19 putative mutants obtained and the three respective controls generated highly reproducible patterns, with multiple discrete bands when they were amplified with the three most polymorphic ISSR primers. Of the 35 amplified bands, 30 corresponded to polymorphic fragments (85.71%) detected with the primers UBC840A, UBC840B and UBC816. All evaluated plants were different respect to respective controls. The present work is the first report of molecular characterization through ISSR markers of Cenchrus ciliaris mutants. This technique revealed a high degree of polymorphism and was successfully used to differentiate these materials. The results suggest that induced mutations may be used as a novel approach within the framework of a genetic improvement program to increase genetic diversity in Cenchrus ciliaris. Further analysis will be necessary to evaluate the suitability of these new genetic resources in terms of biomass production and quality to be incorporated to cattle raising systems.
