Abstract:
This study investigated the dynamics of parasitism, host plant resistance, pathogens, and predation on the demography of wheat stem sawfly, Cephus cinctus Norton (Hymenoptera: Cephidae), developing in susceptible (hollow stem) and resistant (solid stem) wheat hosts. This study is also the first to investigate the prevalence and impact of cannibalism on wheat stem sawfly mortality. Wheat stem sawflies were sampled in two commercial wheat fields over 4 yr from the egg stage through adult emergence, and multiple decrement life tables were constructed and analyzed. Cannibalism, host plant resistance, or unknown factors were the most prevalent factors causing egg mortality. Summer mortality of prediapause larvae ranged from 28 to 84%, mainly due to parasitism by Bracon cephi (Gahan) and Bracon lissogaster Muesebeck, cannibalism, and host plant resistance. Winter mortality ranged from 6 to 54% of the overwintering larvae, mainly due to unknown factors or pathogens. Cannibalism is a major cause of irreplaceable mortality because it is absolute, with only a single survivor in every multiple infested stem. Subsequent to obligate cannibalism, mortality of feeding larvae due to host plant resistance was lower in hollow stem wheat than in solid stem wheat. Mortality from host plant resistance was largely irreplaceable. Irreplaceable mortality due to parasitoids was greater in hollow stem wheat than in solid stem wheat. Host plant resistance due to stem solidness and parasitism in hollow stems cause substantial mortality in populations of actively feeding larvae responsible for all crop losses. Therefore, enhancing these mortality factors is vital to effective integrated pest management of wheat stem sawfly.

Abstract:
Langdon(Dic-3A)-10 line carrying the QTL Qfhs.ndsu-3AS from T. turgidum ssp.dicoccoides that confers Type II resistance to Fusarium head blight (FHB) was crossed with Argentinean durum wheat cultivars. F4 progeny were screened with the microsatellite locus Xgwm2, tightly linked to the Qfhs.ndsu-3A region. Reaction of these plants and parents to FHB was evaluated at 7, 14 and 21 days post-inoculation (dpi) with F. graminearum; severity (% symptomatic spikelets/spike) and AUDPC (area under disease progress curve) were calculated. F4 progeny carrying the resistance allele in heterozygous or in homozygous condition showed significantly lower scab damage at 21 dpi and slower progress of disease than cultivated parents. Our results indicate that the resistance Qfhs.ndsu-3AS has a stable dominance expression in genetic backgrounds of durum cultivars and demonstrate that the linked microsatellite is an effective molecular tool for resistance screening. This work offers valuable information for Qfhs.ndsu-3AS utilization in wheat breeding programs.

Abstract:
The effect of commercial modified celluloses: microcrystalline cellulose, carboxymethyl cellulose, and hydroxypropylmethyl cellulose on bread quality attributes and their potential protective effect with respect to bread staling were analyzed. Two levels of gums were assayed (0.5 and 1.5 g/100 g flour). The best performance was obtained with carboxymethyl cellulose and hydroxypropylmethyl cellulose F 4 M at both levels; these gums led to higher specific volumes and a better crumb texture as measured by texture profile analysis. In general, crumbs were softer, more cohesive, and resilient and exhibited lower chewiness values. Other gums like microcrystalline cellulose and hydroxypropylmethyl cellulose F50 did not improve bread quality on the same extent. Mechanical spectra obtained by dynamic mechanical analysis assays indicated a marked change in molecular mobility when carboxymethyl cellulose was present. Bread staling was evaluated by texture profile analysis, moisture loss, and calorimetric assays. Gums did not avoid retrogradation and even exhibited an accelerating effect, probably due to changes in water retention and migration during storage. However, in most cases, final crumb hardness in samples with hydrocolloids was lower than that in the control sample.

Abstract:
Fusarium proliferatum produces fumonisins B not only on maize but also on diverse crops including wheat. Using a wheat-based medium, the effects of abiotic factors, temperature and water activity (aW), on growth, fumonisin biosynthesis, and expression of FUM genes were compared for three F. proliferatum strains isolated from durum wheat in Argentina. Although all isolates showed similar profiles of growth, the fumonisin production profiles were slightly different. Regarding FUM gene transcriptional control, both FUM8 and FUM19 expression showed similar behavior in all tested conditions. For both genes, expression at 25 °C correlated with fumonisin production, regardless of the aw conditions. However, at 15 °C, these two genes were as highly expressed as at 25 °C although the amounts of toxin were very weak, suggesting that the kinetics of fumonisin production was slowed at 15 °C. This study provides useful baseline data on conditions representing a low or a high risk for contamination of wheat kernels with fumonisins.

Abstract:
Casein glycomacropeptide (CMP) is a 64‐ amino acid peptide found in cheese whey, which is released after κ‐casein specific cleavage by chymosin. CMP lacks aromatic amino acids, a characteristic that makes it usable as a nutritional supplement for people with phenylketonuria. CMP consists of two nonglycosylated isoforms (aCMP A and aCMP B) and its different glycosylated forms (gCMP A and gCMP B). The most predominant carbohydrate of gCMP is N‐acetylneuraminic acid (sialic acid). Here, we developed a CMP purification process based on the affinity of sialic acid for wheat germ agglutinin (WGA). After formation of chitosan beads and adsorption of WGA, the agglutinin was covalently attached with glutaraldehyde. Two matrices with different WGA density were assayed for CMP adsorption. Maximum adsorption capacities were calculated according to the Langmuir model from adsorption isotherms developed at pH 7.0, being 137.0 mg/g for the matrix with the best performance. In CMP reduction from whey, maximum removal percentage was 79% (specifically 33.7% of gCMP A and B, 75.8% of aCMP A, and 93.9% of aCMP B). The CMP was recovered as an aggregate with an overall yield of 64%. Therefore, the matrices developed are promising for CMP purification from cheese whey.

Abstract:
The effect of addition of the nitric oxide donor S-nitrosoglutathione (GSNO) on the Zn nutritional status was evaluated in hydroponically-cultured wheat plants (Triticum aestivum cv. Chinese Spring). Addition of GSNO in Zn-deprived plants did not modify biomass accumulation but accelerated leaf senescence in a mode concomitant with accelerated decrease of Zn allocation to shoots. In well-supplied plants, Zn concentration in both roots and shoots declined due to long term exposure to GSNO. A further evaluation of net Zn uptake rate (ZnNUR) during the recovery of long-term Zn-deprivation unveiled that enhanced Zn-accumulation was partially blocked when GSNO was present in the uptake medium. This effect on uptake was mainly associated with a change of Zn translocation to shoots. Our results suggest a role for GSNO in the modulation of Zn uptake and in root-to-shoot translocation during the transition from deficient to sufficient levels of Zn-supply.

Abstract:
Fast development of seedling leaf area is a relevant trait in order to increase early resource acquisition. The use of semi-dwarf genotypes of wheat has decreased early vigour of modern cultivars. We studied early vigour of 20 cultivars cropped in Argentina, and our main objectives were: (i) to analyse the genotypic variability in early vigour; (ii) to study morphological traits that can be good indicators of early vigour, such as seed mass, leaf width, and specific leaf area; and (iii) to determine whether increased dry mass allocation to roots impacts negatively on early vigour. Experiments with non-size-selected and size-selected seeds were carried out in a greenhouse. A field trial was also conducted in order to test the reliability of the greenhouse results. Seeds mass was the main parameter related to early vigour. However, results from the experiment with seeds selected by size (45-50mg seed-1) showed that seed mass per se only partially explains early vigour, since a significant coefficient of determination was observed between the seedling leaf area of each cultivar in both experiments (i.e. with randomly chosen or size-selected seeds). We observed a high coefficient of determination between net assimilation rate and changes in the ranking of early vigour of the cultivars with time after transplant. Root biomass was positively correlated with leaf area, indicating that the traits were not mutually exclusive. We built simple models by multiple regression to predict early vigour, including some parameters that were easy to measure. Seed mass and leaf width taken together showed better fit than seed mass or leaf width alone. We found a significant coefficient of determination between early vigour in greenhouse and field experiments; thus, screening for early vigour under semi-controlled conditions may be feasible.

The concern regarding toxicity from the presence of deoxynivalenol (DON) in wheat that affects both economy and public health leads to the need to find appropriate detection methods for determining the degree of DON contamination in terms of the equipment available and the speed required for obtaining the incidence. The objective of this study was to compare the performance of two alternative analytical techniques for DON quantification for use in the food industry with a reference technique. Samples of wheat and the commercial by-products were analysed by high-performance liquid chromatography (HPLC) with an ultraviolet detector as the reference method and the results compared with those obtained from a rapid lateral-flow immunochromatographic device (Reveal Q+) and of a Fourier-transform-infrared (FTIR) spectroscopy technique. Pearson's correlation coefficient between the HPLC and Reveal-Q+ data (0.45), although significant (P<0.0003), was lower than that obtained between HPLC and the FTIR method (0.94, P<0.0001). Both methods were considered efficient in quantifying DON levels in wheat-flour samples. This study was aimed at assisting the producers in choosing an appropriate tool for the purpose of analysis and upon consideration of the available equipment.

Abstract:
Paraburkholderia tropica is an endophytic nitrogen-fixing bacterium isolated from the rhizosphere, rhizoplane, and internal tissues of sugarcane and corn plants in different geographical regions. Other plant-growth-promoting abilities, such as phosphate solubilization and antifungal activity, have also been reported for this bacterium. With an aim at investigating the potential use of P. tropica as an inoculant for improving the performance of wheat crop, in this work we evaluated an experimental inoculant formulated with P. tropica MTo-293 with respect to root colonization, the practical aspects of its application, and the effects under field conditions when applied to wheat seeds. Bacterial colonization was monitored by culture dependent techniques and the wheat yield determined by quantifying the total grain production in two different seasons. Rhizoplane and endophytic colonization in wheat roots was achieved efficiently (on average, 8 and 4 log colony-forming units/g fresh weight, respectively) even at relatively low concentrations of viable bacteria in the inoculum under controlled conditions. P. tropica was compatible with a widely used fungicide, maintained viability for 48 h once applied to seeds, and was also able to colonize wheat roots efficiently. Furthermore, we were able to formulate an inoculant that maintained bacterial viability for relatively long time periods. Preliminary field assays were realized, and even though the average yields values for the inoculated treatments remained above the uninoculated ones, no significant effects of inoculation were detected with or without fertilization. The correct physiologic behavior of P. tropica suggests the necessity to continue with field experiments under different conditions.
